ModLoc – Modulated localization microscopy

Abstract

The objective of this project is to develop a new concept in super-resolution optical microscopy.

Modulated Localization Microscopy (ModLoc) represents a radically new approach to encode information from individual emitters. The spatial modulation of the light excitation makes it possible to encode the transverse position of the emitters with a 2-fold gain precision over standard localization techniques. This modulated information is extracted by an original multiplexed homodyne detection taking full advantage of the specificities of super-resolution microscopy without compromising the acquisition time.

ModLoc microscopy brings all the advantages of homodyne detection in super-resolution microscopy in particular this technique is insensitive to all optical aberrations originating either in the sample or in the optical system.

The first results clearly demonstrate an enhance precision along one direction. The goal of the project is to develop a 2D and 3D implementation to reach a sub-5nm precision in all directions at an advanced TRL level.
