Z.O.L.A. 3D – Zernike Optimized Localization Approach in 3D

Abstract

In response to the ever-growing user demand for 3D super resolved imaging, the Institut Pasteur imaging facility (UTechS Photonic BioImaging) intends to duplicate the ZOLA-3D microscopy approach from the Imaging and Modeling research unit. This open optical and computational Single Molecule Localisation Microscopy (SMLM) approach addresses some of the limitations of state-of-the-art commercial systems in terms of axial range and flexibility. Its adaptability to various objectives and samples makes it ideal for a multi-user BSL2 environment. To achieve robust and successful transition of ZOLA we are seeking support to hire a dedicated engineer that will perform meticulous metrology tests, as well as full system characterisation to provide an experimental pipeline for permanent staff to take over. Combined with experienced permanent engineers in the facility and the research unit, this will overcome common challenges in technology transfer and ensure reliable and sustainable user access to this cutting-edge imaging modality.
